When they take him, Santanico tries to go after them, but she is still bound to the temple and cannot leave. In The Take, when the brothers pass the test and Richie swallows the serpent containing Santanico's blood, Carlos reveals his betrayal to her and kidnaps Richie. While in her chambers, she is confronted by Carlos, who tells her not to worry as they'll soon find out where everybody stands.

After Seth goes into the RV after seeing Richie and Santanico, she tells them about the labyrinth and setting her free from the temple. After he tackles "Monica" when she stabs him in the eye, Santanico tells him he's almost there. In Boxman, after Richie awakens and transitioning into a culebra, Santanico tells him to let things go. He eventually gives in and allows her to turn him into a culebra. When Richie wakes up, she tells him what her intentions are and how she became what she is now, that she was just an innocent girl when the tribesmen transformed her into a culebra. In La Conquista, she and Carlos discuss Richie's condition, with her reassuring him things are going the way they should. Freddie then shoots Richie from behind, blood splattering on Santanico and she pushes him against Seth, taking Richie with her. She comes back out to talk to Richie, telling him he belongs there and wanted to show him her what she was on her own terms. She tells them the culebras below them are no longer slaves and begins slaughtering the consiglieres with Carlos after poisoning them. She is then notified of the consiglieres and goes to the feast room to greet them. She is furious with Carlos who told her Freddie was taken care of, and tells him no more mistakes. She then retreats to her chambers and heal the bullet wound she received. She tells Freddie that his fight is over and bites him, then tries to talk to Richie until Seth shoots her. In Pandemonium, she begins to dance and watches as Freddie stabs Richie's hand, threatening both brothers until she turns into her culebra form and jumps on Freddie, then proceeds to start attacking various patrons.
In Place of Dead Roads, she comes on stage after Carlos introduces her to the audience, ready for her show. She warns him to watch out for people with shadows over their hearts and to be wary of both Scott and Jacob.
In Self Contained, she appears to Richie while he is in the RV with his brother and the Fullers to cross the border. In Mistress, she appears to Richie once more, telling him that Monica wants him to set her free and Santanico needs him as well. After some difficulty and Richie taking Monica out of the room, Santanico appears to him again, repeatedly telling him to "cut her". In Blood Runs Thick, she appears to Richie as he begins opening the bank safe, telling him to "set her free". They begin biting and when a shaman casts a spell, a snake crawls inside her mouth. In Pilot, she is seen in a flashback running away from many men she then falls into a pit of snakes.

It was decided that because of her refusal to sacrifice to the Lords, she would be given a deep hunger for blood, one severe enough to make it her lifeline. But the priest found her again, and she was punished for not obeying them. As a result, she ran, desperate to be free. This was done as she refused to spill any more blood in the temple she was kept at for the Lords. One snake slithered into her mouth (something that is shown later to be the original cause of turning someone into a culebra). Once they caught her, they threw her into a pit of snakes, which bit her, causing her to scream. In ancient Mesoamerican times, she attempted to escape from a group of men that were imprisoning her due to her alleged connection to the Gods. Kisa's family was killed by Amancio Malvado when she was younger.
